Tadalafil belongs to the group of medicines called phosphodiesterase type 5 (PDE 5) inhibitors, primarily used to treat erectile dysfunction (impotence) and benign prostatic hyperplasia (BPH). Erectile dysfunction is the inability to keep a hard and erect penis suitable for sexual activity. Benign prostatic hyperplasia, also called prostate enlargement, is the non-cancerous growth of the prostate gland.
Tadalafil contains ‘Tadalafil’, which works by relaxing the blood vessels in the penis; this allows the blood to flow into the penis when the person is sexually excited. Thereby, it helps to treat erectile dysfunction. Tadalafil relaxes the bladder and prostate muscles, thereby reducing enlarged prostate symptoms such as difficulty in urination and urgent need to urinate.
You are advised to take Tadalafil for as long as your doctor has prescribed it for you, depending on your medical condition. Erectile dysfunction (ED) is defined as the inability to achieve or maintain an erection sufficient for satisfactory sexual intercourse. The prevalence of ED in males is estimated at 1–2% of the world’s population and this prevalence is believed to be increasing as a result of aging and an increase in the incidence of sexual problems.1
Erectile dysfunction (ED) can cause psychological disorders, such as depression and anxiety, and can lead to the onset of ED. The prevalence of ED in men in India is quite high, and it is estimated that 80–90% of men will experience sexual dysfunction at some point in their lifetime.2,3 The prevalence of ED in men in Pakistan is estimated to be between 2–10% and 20% for men in general population, and this percentage increases with age.4
The prevalence of ED in Pakistan is estimated to be around 2% in general population.5 The prevalence of ED in men in India is estimated to be between 2% and 15% in Pakistan.6 The prevalence of ED in men in the United Kingdom is estimated to be between 10% and 20% in the UK.7 The prevalence of ED in men in the United States is estimated to be around 5% in men in general population.8
The prevalence of ED in men in the UK is estimated to be between 4% and 15%.9 The prevalence of ED in men in the US is estimated to be between 7% and 15%.9
ED treatment is recommended for men with ED. The recommended dosage of tadalafil in men with ED is 1 mg taken once daily, which is suitable for patients with mild-to-moderate erectile dysfunction.10
Tadalafil is a phosphodiesterase type 5 (PDE5) inhibitor. It belongs to a class of drugs known as PDE5 inhibitors. Tadalafil inhibits the phosphodiesterase type 5 enzyme, which is responsible for the degradation of cyclic guanosine monophosphate (cGMP). cGMP is a chemical messenger that plays a key role in controlling smooth muscle relaxation and blood flow in the penile region.11 Tadalafil is rapidly absorbed in the body, and it reaches peak plasma concentration within 1–2 h of oral administration, and it has a half-life of approximately 36 h.12,13
Tadalafil is the active ingredient in the pharmaceutical formulation, which is indicated for the treatment of erectile dysfunction.13 The recommended dosage and administration route of tadalafil in men with ED is as follows:13
This study was conducted to evaluate the effect of tadalafil in combination with dapoxetine in patients with ED. The study was conducted in two phases; a phase I, comprised 15 clinical trials in men with ED and 15 control trials in men without ED. The results of the phase II trial were evaluated and compared with a control trial, which was not designed for this purpose.
A total of 806 men with ED were included in this study, with the mean age of 45.57 ± 9.53, and the mean age range was between 26.62 and 32.66 years. The study was conducted at an average of 3.20 ± 1.14 h. The mean concentration of tadalafil in the study group was 2.33 ± 1.20 mg/mL, and the mean concentration in the control group was 1.83 ± 0.70 mg/mL. Tadalafil in combination with dapoxetine was well tolerated in both groups, with no significant side effects, and no adverse drug effects were observed in the control group. In addition, there were no significant differences in the demographic, clinical, or laboratory parameters between tadalafil and dapoxetine.
The mean age of the patients was 46.13 ± 7.53 years, and the mean age range was between 22.26 and 34.82 years, and the mean age of patients was between 23.44 and 35.65 years, respectively. The mean plasma concentration of tadalafil in the study group was 1.33 ± 0.13 mg/mL, and the mean plasma concentration in the control group was 1.84 ± 0.50 mg/mL. The mean concentration of tadalafil in the control group was 2.33 ± 0.28 mg/mL, and the mean concentration in the study group was 3.33 ± 1.13 mg/mL.
